TY Recipes

LB Broth/LB Agar

1 Liter

Tryptone 10g

Yeast Extract 5g

NaCl 5g

Agar (for plates) 15g

ddH2O to 1 Liter

Wet TY Plates (aka TY agar or molten TY or soft TY agar)

Prepare LB agar as indicated above.

-after autoclaving add the following:

10.0 ml 1M MgSO4

1.0 ml MnSO4

Pour plates and store @ 4C immediately after agar solidifies.

TY Broth

100ml

to 100ml sterile LB Broth add:

1.0 ml 1M MgSO4

0.1 ml 0.1M MnSO4

Store RT

Soft TY Agar Bottles

100 ml

Prepare broth as above (it’s easiest to prepare 1 L LB broth and aliquot 100 ml into 10 bottles)

Add .5 g agar to each 100 ml bottle

After autoclaving, add the following to each bottle:

1 mL 1M MgSO4

100 uL 0.1 M MnSO4
